PIINP大鼠Ⅱ型前膠原氨(an)基(ji)端原肽Elisa試劑(ji)盒 實驗(yan)目的(de):本(ben)試劑盒用于定量測定大鼠血(xue)清,血(xue)漿及(ji)相關液體樣本(ben)中Ⅱ型(xing)前(qian)膠原氨基端原肽(PIINP)的(de)含(han)量。
本試劑(ji)僅(jin)供研(yan)究使(shi)用 不得用于體內(nei)診斷(duan)
樣本(ben)處理及(ji)要(yao)求:
1. 血清:室(shi)溫血液自然凝固10-20分(fen)鐘,離心20分(fen)鐘左右(you)(2000-3000轉/分(fen))。仔細(xi)收集上清,保(bao)存過程中如出現沉淀(dian),應再次(ci)離心。
2. 血漿:應(ying)根據標(biao)本的要(yao)求選(xuan)擇(ze)EDTA或檸檬酸鈉作(zuo)為(wei)抗凝劑,混合10-20分(fen)鐘后,離(li)心20分(fen)鐘左右(you)(2000-3000轉/分(fen))。仔細收集(ji)上清,保存(cun)過程中如(ru)有沉淀形成,應(ying)該再(zai)次離(li)心。
3. 尿液(ye):用無菌(jun)管收集,離心20分鐘左右(2000-3000轉(zhuan)/分)。仔細收集上清,保存過(guo)程中如有(you)沉淀形成,應再次離心。胸腹水(shui)、腦(nao)脊液(ye)參照實行(xing)。
4. 細(xi)胞(bao)培養上清:檢(jian)測(ce)分(fen)泌性的成份時,用無菌(jun)管(guan)收集。離心(xin)20分(fen)鐘(zhong)左右(you)(2000-3000轉/分(fen))。仔細(xi)收集上清。檢(jian)測(ce)細(xi)胞(bao)內的成份時,用PBS(PH7.2-7.4)稀釋細(xi)胞(bao)懸液,細(xi)胞(bao)濃度達到(dao)100萬/ml左右(you)。通過(guo)反復凍融,以使細(xi)胞(bao)破(po)壞并放出細(xi)胞(bao)內成份。離心(xin)20分(fen)鐘(zhong)左右(you)(2000-3000轉/分(fen))。仔細(xi)收集上清。保存過(guo)程中(zhong)如有沉淀形成,應再次離心(xin)。
5. 組織(zhi)標(biao)本:切割標(biao)本后,稱取重量。加入(ru)一(yi)(yi)定量的PBS,PH7.4。用(yong)液(ye)氮迅速冷凍保(bao)存備(bei)用(yong)。標(biao)本融化后仍然(ran)保(bao)持2-8℃的溫度。加入(ru)一(yi)(yi)定量的PBS(PH7.4),用(yong)手工(gong)或勻(yun)漿器將標(biao)本勻(yun)漿充(chong)分(fen)。離心20分(fen)鐘左右(2000-3000轉/分(fen))。仔(zi)細收集上清(qing)。分(fen)裝后一(yi)(yi)份待檢測,其余(yu)冷凍備(bei)用(yong)。
6. 標本(ben)采集后盡早(zao)進(jin)行提取,提取按(an)相關文(wen)獻(xian)進(jin)行,提取后應(ying)盡快進(jin)行實驗(yan)。若不能馬(ma)上進(jin)行試驗(yan),可將標(biao)本放于-20℃保存,但應(ying)避免反復凍(dong)融.
7. 不能檢(jian)測(ce)含(han)NaN3的樣品,因NaN3抑制辣根(gen)過氧化物酶的(HRP)活性。
實驗原理(li):
本(ben)試劑盒應用(yong)雙抗(kang)(kang)體夾(jia)心(xin)法測(ce)定標本(ben)中大(da)鼠Ⅱ型前膠(jiao)(jiao)(jiao)原(yuan)(yuan)(yuan)(yuan)氨(an)基端原(yuan)(yuan)(yuan)(yuan)肽(tai)(PIINP)的(de)(de)(de)(de)水平。用(yong)純化的(de)(de)(de)(de)大(da)鼠Ⅱ型前膠(jiao)(jiao)(jiao)原(yuan)(yuan)(yuan)(yuan)氨(an)基端原(yuan)(yuan)(yuan)(yuan)肽(tai)(PIINP)抗(kang)(kang)體包(bao)被微孔板,制成(cheng)固相抗(kang)(kang)體,往包(bao)被單抗(kang)(kang)的(de)(de)(de)(de)微孔中依(yi)次加入大(da)鼠Ⅱ型前膠(jiao)(jiao)(jiao)原(yuan)(yuan)(yuan)(yuan)氨(an)基端原(yuan)(yuan)(yuan)(yuan)肽(tai)(PIINP)再與HRP標記的(de)(de)(de)(de)Ⅱ型前膠(jiao)(jiao)(jiao)原(yuan)(yuan)(yuan)(yuan)氨(an)基端原(yuan)(yuan)(yuan)(yuan)肽(tai)(PIINP)抗(kang)(kang)體結合(he),形成(cheng)抗(kang)(kang)體-抗(kang)(kang)原(yuan)(yuan)(yuan)(yuan)-酶標抗(kang)(kang)體復合(he)物(wu),經過(guo)*洗滌后加底(di)物(wu)TMB顯色(se)(se)。TMB在HRP酶的(de)(de)(de)(de)催化下轉化成(cheng)藍色(se)(se),并(bing)在酸的(de)(de)(de)(de)作(zuo)用(yong)下轉化成(cheng)zui終的(de)(de)(de)(de)黃色(se)(se)。顏色(se)(se)的(de)(de)(de)(de)深淺和(he)樣(yang)品中的(de)(de)(de)(de)Ⅱ型前膠(jiao)(jiao)(jiao)原(yuan)(yuan)(yuan)(yuan)氨(an)基端原(yuan)(yuan)(yuan)(yuan)肽(tai)(PIINP)呈正相關。用(yong)酶標儀在450nm波長下測(ce)定吸光度(du)(OD值),通過(guo)標準曲線(xian)計算樣(yang)品中大(da)鼠Ⅱ型前膠(jiao)(jiao)(jiao)原(yuan)(yuan)(yuan)(yuan)氨(an)基端原(yuan)(yuan)(yuan)(yuan)肽(tai)(PIINP)濃度(du)。

操作步驟:

2.加樣(yang):分別(bie)設空白孔(kong)(空白對(dui)照孔(kong)不加樣(yang)品及酶(mei)(mei)標試劑,其(qi)余(yu)各步操(cao)作(zuo)相同)、待(dai)測(ce)(ce)(ce)樣(yang)品孔(kong)。在酶(mei)(mei)標包被(bei)板上待(dai)測(ce)(ce)(ce)樣(yang)品孔(kong)中先加樣(yang)品稀(xi)釋(shi)液40μl,然后再加待(dai)測(ce)(ce)(ce)樣(yang)品10μl(樣(yang)品zui終稀(xi)釋(shi)度為5倍)。加樣(yang)將樣(yang)品加于(yu)酶(mei)(mei)標板孔(kong)底部,盡量不觸及孔(kong)壁,輕輕晃(huang)動混勻。
3.溫育:用封板膜(mo)封板后置(zhi)37℃溫育(yu)30分鐘。
4.配液:將(jiang)30(48T的20倍(bei))倍(bei)濃縮洗滌(di)液用(yong)蒸(zheng)餾(liu)水30(48T的20倍(bei))倍(bei)稀釋后備用(yong)。
5.洗滌:小(xiao)心(xin)揭掉(diao)封板膜,棄(qi)去液體,甩干(gan),每孔加滿洗滌液,靜置30秒后(hou)棄(qi)去,如此重復5次,拍干(gan)。
6.加酶:每孔加入(ru)酶標試劑50μl,空白孔除外。
7.溫(wen)育:操作同3。
8.洗滌(di):操作同(tong)5。
9.顯(xian)色:每孔先加入顯(xian)色劑(ji)A50μl,再加入顯(xian)色劑(ji)B50μl,輕輕震蕩混勻,37℃避光顯色(se)15分鐘.
10.終(zhong)止:每(mei)孔加終(zhong)止液50μl,終(zhong)止反應(此時藍(lan)色(se)立轉黃色(se))。
11.測(ce)定:以(yi)空(kong)白空(kong)調零,450nm波長依序測(ce)量各孔的吸光度(OD值)。 測(ce)定應在加(jia)終止液后15分鐘以(yi)內進行。
計算:
以(yi)標(biao)準物的(de)濃度為橫坐(zuo)標(biao),OD值為縱坐(zuo)標(biao),在坐(zuo)標(biao)紙上繪出標(biao)準曲線,根據(ju)樣品的(de)OD值(zhi)由(you)標(biao)準曲線查出相(xiang)應的(de)濃度;再乘以稀釋(shi)倍數;或用標(biao)準物(wu)的(de)濃度與OD值(zhi)計算(suan)出標(biao)準曲(qu)線的直線回歸方(fang)程(cheng)式,將樣(yang)品的OD值代(dai)入(ru)方(fang)程(cheng)式,計算出樣(yang)品濃度,再(zai)乘以(yi)稀釋倍數(shu),即為樣品的實際濃(nong)度。
