本試(shi)劑僅供研究使用       

目的(de):本(ben)試劑盒(he)用于測定大鼠(shu)血清,血漿及相關(guan)液體(ti)樣本(ben)中不對稱二(er)甲基精(jing)氨酸(ADMA)的(de)含量。

實驗原理:

  本試劑盒應(ying)用(yong)(yong)雙抗(kang)體夾心法測定標本中大(da)鼠不(bu)(bu)對稱(cheng)(cheng)二甲(jia)基精(jing)(jing)氨(an)(an)酸(ADMA)水平。用(yong)(yong)純(chun)化(hua)(hua)的大(da)鼠不(bu)(bu)對稱(cheng)(cheng)二甲(jia)基精(jing)(jing)氨(an)(an)酸(ADMA)抗(kang)體包被(bei)微(wei)孔板,制成(cheng)(cheng)固相抗(kang)體,往包被(bei)單抗(kang)的微(wei)孔中依次(ci)加(jia)入(ru)不(bu)(bu)對稱(cheng)(cheng)二甲(jia)基精(jing)(jing)氨(an)(an)酸(ADMA),再與(yu)HRP標記的羊(yang)抗(kang)鼠抗(kang)體結(jie)合(he),形成(cheng)(cheng)抗(kang)體-抗(kang)原-酶(mei)標抗(kang)體復合(he)物(wu),經(jing)過(guo)*洗滌后加(jia)底物(wu)TMB顯色(se)。TMB在HRP酶(mei)的催化(hua)(hua)下轉(zhuan)化(hua)(hua)成(cheng)(cheng)藍色(se),并在酸的作用(yong)(yong)下轉(zhuan)化(hua)(hua)成(cheng)(cheng)zui終的黃色(se)。顏色(se)的深淺和樣品中的不(bu)(bu)對稱(cheng)(cheng)二甲(jia)基精(jing)(jing)氨(an)(an)酸(ADMA)呈(cheng)正相關(guan)。用(yong)(yong)酶(mei)標儀在450nm波長下測定吸(xi)光度(OD值),通(tong)過(guo)標準曲線計(ji)算樣品中大(da)鼠不(bu)(bu)對稱(cheng)(cheng)二甲(jia)基精(jing)(jing)氨(an)(an)酸(ADMA)濃度。

樣本處理及要求:

1. 血清(qing):室(shi)溫血液自(zi)然凝固10-20分鐘,離(li)心20分鐘左(zuo)右(2000-3000轉/分)。仔細收集(ji)上清(qing),保存過(guo)程(cheng)中如出現(xian)沉淀,應再次離(li)心。

2. 血漿:應(ying)(ying)根(gen)據標本(ben)的要求選擇EDTA或檸(ning)檬酸鈉(na)作為抗凝劑,混合10-20分(fen)鐘后,離心(xin)20分(fen)鐘左右(you)(2000-3000轉/分(fen))。仔細(xi)收集(ji)上清,保存過程中如有沉(chen)淀形成(cheng),應(ying)(ying)該(gai)再(zai)次離心(xin)。

3. 尿液(ye):用無菌管收集(ji),離心20分鐘(zhong)左右(2000-3000轉/分)。仔細收集(ji)上清,保存過程(cheng)中如有沉(chen)淀形成,應再次離心。胸腹水、腦脊液(ye)參照實行。

4. 細(xi)(xi)胞(bao)培養上清(qing):檢測(ce)(ce)分泌性的成份(fen)時(shi),用無菌(jun)管收集(ji)。離(li)(li)心20分鐘(zhong)左右(2000-3000轉/分)。仔細(xi)(xi)收集(ji)上清(qing)。檢測(ce)(ce)細(xi)(xi)胞(bao)內的成份(fen)時(shi),用PBS(PH7.2-7.4)稀釋細(xi)(xi)胞(bao)懸液,細(xi)(xi)胞(bao)濃(nong)度(du)達到(dao)100萬(wan)/ml左右。通過反復凍融(rong),以使細(xi)(xi)胞(bao)破壞(huai)并(bing)放出細(xi)(xi)胞(bao)內成份(fen)。離(li)(li)心20分鐘(zhong)左右(2000-3000轉/分)。仔細(xi)(xi)收集(ji)上清(qing)。保(bao)存過程(cheng)中(zhong)如有沉淀形(xing)成,應再次離(li)(li)心。

5. 組織標本(ben):切割標本(ben)后(hou),稱取重(zhong)量(liang)(liang)。加入一(yi)定(ding)量(liang)(liang)的(de)(de)PBS,PH7.4。用(yong)液氮(dan)迅速冷(leng)凍保存(cun)備用(yong)。標本(ben)融(rong)化后(hou)仍然保持2-8℃的(de)(de)溫度。加入一(yi)定(ding)量(liang)(liang)的(de)(de)PBS(PH7.4),用(yong)手工或勻漿器(qi)將標本(ben)勻漿充分(fen)(fen)。離心(xin)20分(fen)(fen)鐘(zhong)左右(2000-3000轉/分(fen)(fen))。仔細(xi)收集(ji)上清(qing)。分(fen)(fen)裝后(hou)一(yi)份(fen)待(dai)檢測,其余冷(leng)凍備用(yong)。

6. 標本采集后盡早進(jin)行提(ti)取(qu),提(ti)取(qu)按(an)相關文(wen)獻進(jin)行,提(ti)取(qu)后應盡快進(jin)行實驗。若不能馬(ma)上進(jin)行試驗,可將標本放于-20℃保存,但應避免(mian)反復凍融.

7. 不能檢測含(han)NaN3的(de)樣(yang)品(pin),因NaN3抑制辣根過氧化物酶的(HRP)活性(xing)。


操作步驟(zou):

  1. 標準(zhun)(zhun)(zhun)品(pin)(pin)(pin)的稀釋(shi)(shi)(shi)與加(jia)(jia)樣:在酶標包被板上設(she)標準(zhun)(zhun)(zhun)品(pin)(pin)(pin)孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)10孔(kong)(kong)(kong)(kong)(kong)(kong)(kong),在*、第(di)(di)(di)二孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong)分(fen)別(bie)(bie)加(jia)(jia)標準(zhun)(zhun)(zhun)品(pin)(pin)(pin)100μl,然后(hou)(hou)在*、第(di)(di)(di)二孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong)加(jia)(jia)標準(zhun)(zhun)(zhun)品(pin)(pin)(pin)稀釋(shi)(shi)(shi)液(ye)50μl,混(hun)(hun)勻(yun)(yun);然后(hou)(hou)從*孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)、第(di)(di)(di)二孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong)各(ge)(ge)取100μl分(fen)別(bie)(bie)加(jia)(jia)到第(di)(di)(di)三(san)孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)和(he)第(di)(di)(di)四孔(kong)(kong)(kong)(kong)(kong)(kong)(kong),再(zai)(zai)在第(di)(di)(di)三(san)、第(di)(di)(di)四孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)分(fen)別(bie)(bie)加(jia)(jia)標準(zhun)(zhun)(zhun)品(pin)(pin)(pin)稀釋(shi)(shi)(shi)液(ye)50μl,混(hun)(hun)勻(yun)(yun);然后(hou)(hou)在第(di)(di)(di)三(san)孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)和(he)第(di)(di)(di)四孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong)先(xian)各(ge)(ge)取50μl棄掉,再(zai)(zai)各(ge)(ge)取50μl分(fen)別(bie)(bie)加(jia)(jia)到第(di)(di)(di)五、第(di)(di)(di)六孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong),再(zai)(zai)在第(di)(di)(di)五、第(di)(di)(di)六孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong)分(fen)別(bie)(bie)加(jia)(jia)標準(zhun)(zhun)(zhun)品(pin)(pin)(pin)稀釋(shi)(shi)(shi)液(ye)50ul,混(hun)(hun)勻(yun)(yun);混(hun)(hun)勻(yun)(yun)后(hou)(hou)從第(di)(di)(di)五、第(di)(di)(di)六孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong)各(ge)(ge)取50μl分(fen)別(bie)(bie)加(jia)(jia)到第(di)(di)(di)七(qi)、第(di)(di)(di)八(ba)孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong),再(zai)(zai)在第(di)(di)(di)七(qi)、第(di)(di)(di)八(ba)孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong)分(fen)別(bie)(bie)加(jia)(jia)標準(zhun)(zhun)(zhun)品(pin)(pin)(pin)稀釋(shi)(shi)(shi)液(ye)50μl,混(hun)(hun)勻(yun)(yun)后(hou)(hou)從第(di)(di)(di)七(qi)、第(di)(di)(di)八(ba)孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong)分(fen)別(bie)(bie)取50μl加(jia)(jia)到第(di)(di)(di)九、第(di)(di)(di)十(shi)孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong),再(zai)(zai)在第(di)(di)(di)九第(di)(di)(di)十(shi)孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)分(fen)別(bie)(bie)加(jia)(jia)標準(zhun)(zhun)(zhun)品(pin)(pin)(pin)稀釋(shi)(shi)(shi)液(ye)50μl,混(hun)(hun)勻(yun)(yun)后(hou)(hou)從第(di)(di)(di)九第(di)(di)(di)十(shi)孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)中(zhong)(zhong)各(ge)(ge)取50μl棄掉。(稀釋(shi)(shi)(shi)后(hou)(hou)各(ge)(ge)孔(kong)(kong)(kong)(kong)(kong)(kong)(kong)加(jia)(jia)樣量都(dou)為50μl,濃度分(fen)別(bie)(bie)為600μmol/L,400μmol/L,200μmol/L,100μmol/L,50μmol/L)。
  2. 加(jia)樣(yang)(yang)(yang):分別設(she)空(kong)白(bai)孔(kong)(空(kong)白(bai)對照孔(kong)不加(jia)樣(yang)(yang)(yang)品(pin)及酶(mei)標試劑,其(qi)余各步操作相(xiang)同)、待測(ce)(ce)樣(yang)(yang)(yang)品(pin)孔(kong)。在酶(mei)標包被板上待測(ce)(ce)樣(yang)(yang)(yang)品(pin)孔(kong)中先(xian)加(jia)樣(yang)(yang)(yang)品(pin)稀(xi)釋(shi)液40μl,然(ran)后(hou)再加(jia)待測(ce)(ce)樣(yang)(yang)(yang)品(pin)10μl(樣(yang)(yang)(yang)品(pin)zui終稀(xi)釋(shi)度為5倍)。加(jia)樣(yang)(yang)(yang)將樣(yang)(yang)(yang)品(pin)加(jia)于酶(mei)標板孔(kong)底部,盡量不觸及孔(kong)壁,輕輕晃(huang)動混勻。
  3. 溫育:用封(feng)板(ban)膜(mo)封(feng)板(ban)后(hou)置37℃溫育30分(fen)鐘。
  4. 配液:將30(48T的20倍(bei))倍(bei)濃縮洗滌液用蒸餾水30(48T的20倍(bei))倍(bei)稀釋(shi)后(hou)備(bei)用。
  5. 洗滌:小心揭(jie)掉封板膜,棄(qi)去液體,甩干,每孔(kong)加滿洗滌液,靜(jing)置30秒后棄(qi)去,如此重復(fu)5次(ci),拍干。
  6. 加(jia)酶:每孔加(jia)入酶標試劑50μl,空白孔除外。
  7. 溫育(yu):操作同3。
  8. 洗滌(di):操作(zuo)同(tong)5。
  9. 顯色:每(mei)孔先(xian)加(jia)入顯色劑A50μl,再加(jia)入顯色劑B50μl,輕輕震蕩混勻,37℃避光顯色15分鐘.
  10. 終(zhong)止:每(mei)孔加(jia)終(zhong)止液50μl,終(zhong)止反應(此時藍色(se)立轉(zhuan)黃色(se))。
  11. 測定:以(yi)(yi)空(kong)白空(kong)調零,450nm波長(chang)依序測量各孔的吸光度(OD值)。 測定應在(zai)加終止液后15分(fen)鐘以(yi)(yi)內進行。


注(zhu)意事(shi)項(xiang):

  1. 試(shi)劑(ji)盒從(cong)冷藏環境(jing)中(zhong)取出應(ying)在室溫平衡15-30分鐘后(hou)方(fang)可(ke)使用,酶標包被(bei)板開封后(hou)如未用完(wan),板條(tiao)應(ying)裝入密封袋(dai)中(zhong)保存。
  2. 濃洗滌液(ye)可(ke)能會(hui)有結晶析出(chu),稀釋時可(ke)在水浴中加溫助溶,洗滌時不(bu)影響結果。
  3. 各步加(jia)(jia)樣均應使(shi)用加(jia)(jia)樣器,并經(jing)常校對其準確(que)性,以(yi)避免試驗(yan)誤差。一次(ci)加(jia)(jia)樣時(shi)間控制(zhi)在5分鐘(zhong)內,如(ru)標本數量多,推(tui)薦使(shi)用排(pai)槍加(jia)(jia)樣。
  4. 請每次測定的同時(shi)做(zuo)標(biao)準(zhun)曲線,做(zuo)復孔。如標(biao)本(ben)中待測物質含量過高(樣本(ben)OD值(zhi)大于標(biao)準(zhun)品孔*孔的OD值(zhi)),請先用樣品稀釋(shi)液稀釋(shi)一定倍數(n倍)后再測定,計算時(shi)請zui后乘以總稀釋(shi)倍數(×n×5)。
  5. 封(feng)板膜只限一次性使用,以避免交叉污染。
  6. 底物請避光保存。
  7. 嚴格(ge)按照說明書的操作進行,試驗結果判定(ding)必須以(yi)酶標(biao)儀讀數為準.
  8. 所有樣品,洗(xi)滌(di)液和(he)各種(zhong)廢(fei)棄物(wu)都應(ying)按傳染物(wu)處理。
  9. 本(ben)試(shi)劑不同批號組分不得混用。

10. 如與英(ying)文說明(ming)書(shu)有異(yi),以英(ying)文說明(ming)書(shu)為準。


計算:

以標準物的濃度為橫(heng)坐標,OD值為縱(zong)坐標,   

在(zai)坐標紙上繪出(chu)標準曲線,根(gen)據樣品的OD     

值(zhi)由(you)標準曲線查出相應(ying)的濃度(du);再乘以稀(xi)釋     

倍數;或(huo)用(yong)標(biao)(biao)準物的(de)濃(nong)度(du)與OD值計(ji)算出標(biao)(biao)     

準曲線的直線回歸方程式(shi),將樣品的OD值     

代入(ru)方程式,計算出樣(yang)品濃度(du),再乘以稀(xi)釋(shi)     

倍數,即為樣品的實際(ji)濃度。              ;
