1.細(xi)胞形態(tai)觀察。細(xi)胞膜(mo)是(shi)每天(tian)必(bi)須觀察的(de),看是(shi)否清(qing)晰,細(xi)胞的(de)死亡(wang)往(wang)(wang)往(wang)(wang)是(shi)從細(xi)胞膜(mo)的(de)破裂開始的(de),早(zao)期的(de)表現就是(shi)細(xi)胞膜(mo)某一(yi)點出現欠完整或者毛發(fa)影。
2.細(xi)胞(bao)往(wang)往(wang)會有“抱團(tuan)(tuan)”現象,抱團(tuan)(tuan)生長或(huo)死亡,個人認為應該勤(qin)換液或(huo)傳代(dai),盡量避 免細(xi)胞(bao)抱團(tuan)(tuan)。
3.細胞生(sheng)長迅速,強烈建議用(yong)(yong)較大的培養瓶養,用(yong)(yong)小瓶養的話,換液(ye)不及時,很容易(yi)死 亡(wang)。細胞的生(sheng)長速度(du)一般為2天左右就(jiu)需要(yao)傳代,并且(qie)一般為一傳三,或(huo)一傳四。
4.培養(yang)(yang)液:1640|DMEM|MEM|IMDM|a-MEM培養(yang)(yang)液,1L一般加碳酸(suan)氫(qing)鈉(na)2g,并加雙抗,胎牛血清(qing)濃度*為12%,zui低不(bu)低于(yu)10%,有些細胞對(dui)胎牛血清(qing)高度依賴(lai),比如WRO細胞,HT-22細胞,HA-VSMC細(xi)胞,NK-92細(xi)胞等。培養液(ye)PH應(ying)調為7.20-7.40之間。
5.防污(wu)染(ran)。L-02細胞生長迅速(su),較少發(fa)生污(wu)染(ran),但常規措施應該做好。比如無(wu)菌(jun)操作,酒精(jing) 消毒等,培養瓶口建(jian)議過(guo)(guo)火,包括瓶蓋(gai),應過(guo)(guo)火4秒左右。
6.凍存。-20一小時,-80次(ci)夜,液(ye)氮長(chang)(chang)期保存。強烈建(jian)議液(ye)氮長(chang)(chang)期保存,盡量不要在(zai)-80長(chang)(chang)期 保存,死亡率很高。
7.復蘇。調節水(shui)浴箱溫度(du)為(wei)42℃,并提前在(zai)試(shi)(shi)管(guan)內(nei)放置(zhi)10倍培養(yang)液,使(shi)培養(yang)液溫度(du)與室溫一 致。1分鐘內(nei)融(rong)化細胞(bao)(禁止搖(yao)晃(huang)凍存管(guan)),移取至試(shi)(shi)管(guan)內(nei),動作(zuo)要輕,從試(shi)(shi)管(guan)管(guan)底開始慢慢上(shang)提 移液管(guan),使(shi)細胞(bao)在(zai)培養(yang)液內(nei)分布均勻,離(li)心,洗(xi)2次。
