1. 從(cong)增殖期(qi)(qi)到形成致密的單層(ceng)細胞以(yi)前的培養細胞都可以(yi)用(yong)于(yu)凍(dong)存,但(dan)為對(dui)數生長期(qi)(qi)細胞。在凍(dong)存前一(yi)天換(huan)一(yi)次培養液。
2. 將凍存管放入(ru)液(ye)氮容器或從中取出時,要(yao)做好防護工作,以免凍傷。
3. 凍存和復(fu)蘇(su)用新配制的培(pei)養(yang)液。

當(dang)細胞冷到零度以(yi)下,可(ke)以(yi)產生(sheng)以(yi)下變化:細胞器(qi)脫水,細胞中可(ke)溶(rong)性物質濃度升(sheng)高(gao),并在細胞內形(xing)成(cheng)冰(bing)晶。
如(ru)果緩慢(man)冷(leng)凍,可使細胞(bao)逐(zhu)步(bu)脫水,細胞(bao)內不致(zhi)產生大的冰晶(jing);相反,結(jie)晶(jing)就大,大結(jie)晶(jing)會造成(cheng)細胞(bao)膜、綱胞(bao)器的損傷(shang)和破裂。復蘇過(guo)程應快融,目的是防止小(xiao)冰晶(jing)形成(cheng)大冰晶(jing),即冰晶(jing)的重(zhong)結(jie)晶(jing)。
